As a wood-inhabiting parasite, it is taxonomically related to other significant forest pathogens, making it a subject of international concern in the context of plant health and forest pathology.

The primary hosts for this species are coniferous trees, particularly those within the genus Pinus. The nematode populates the wood tissues, primarily invading the resin canals and xylem parenchyma, where it feeds and multiplies, significantly weakening the physiological state of the host plant.

The life cycle of Bursaphelenchus rainulfi is characterized by a dispersal phase mediated by insect vectors, specifically longhorn beetles of the genus Monochamus. The nematode larvae aggregate in the tracheae of the beetles during the insect's pupal stage and are transmitted to healthy host trees when the beetles engage in maturation feeding on young shoots.
