Unlike some other smut fungi, it is characterized by the unique morphology of its teliospores, which feature a distinct reticulated and rugose surface, giving the species its scientific name.

The host range of Tilletia rugispora primarily includes various species of the Poaceae family. The fungus invades the developing ovary, consuming the nutritious tissue and replacing the entire grain with a mass of dark, powdery spores encased in a persistent pericarp. This process effectively sterilizes the plant, resulting in significant yield loss as the grains fail to develop normally.

Symptoms of the infection become most apparent as the crop reaches maturity. Infected heads often appear slightly smaller or deformed, with glumes that splay outward due to the increased size of the spore-filled kernels.
