The biology of this species is exclusively linked to animal hosts, specifically rodents such as the cotton rat (Sigmodon hispidus). The life cycle involves blood-feeding mites that serve as intermediate hosts, transmitting larvae between vertebrate hosts.

Unlike plant-parasitic nematodes, which cause severe economic losses in agriculture, Litomosoides sigmodontis does not feed on plant roots, vascular systems, or foliage. It is strictly a zoonotic parasite used in laboratory research for studying immunological responses in mammals.
