Its primary application is found in veterinary medicine and apiculture, where it acts as a reliable tool for controlling ectoparasites, most notably the Varroa destructor mite that poses a significant threat to honey bee populations worldwide.

The mechanism of action of coumaphos is centered on the irreversible inhibition of the enzyme acetylcholinesterase within the nervous system of the target pest. By preventing the breakdown of the neurotransmitter acetylcholine, the chemical triggers continuous nerve impulse stimulation, eventually leading to paralysis and the death of the mite. This systemic disruption is highly effective at specific contact concentrations.

In practice, coumaphos is commonly formulated into plastic strips that are inserted into honey bee hives. As bees move throughout the hive, they brush against the strips, distributing the active ingredient. This mechanism ensures that the treatment reaches mites attached to adult bees as well as those hiding within the capped brood cells, providing comprehensive control of the infestation.

Applying this substance requires careful adherence to regional agricultural and veterinary regulations. Timing is critical to prevent the accumulation of chemical residues in honey and wax. Beekeepers are advised to avoid usage during major honey flow periods and to rotate different classes of acaricides to prevent the development of mite resistance, which is a known challenge in modern apiary management.

Safety precautions must be strictly observed, as coumaphos is classified as toxic to humans and other mammals. Handlers should wear protective gloves during application and avoid ingestion, inhalation, or skin contact.
