The primary hosts for Xiphinema diffusum include a wide range of perennial crops such as grapevines, deciduous fruit trees, and various ornamental species. These nematodes prefer the rhizosphere of plants with extensive root systems, where they can establish stable feeding sites and multiply over several growing seasons.

The biology of this species is centered on its ectoparasitic behavior. Using a long, needle-like stylet, the nematode penetrates the root cells, injecting saliva that facilitates the digestion of cell contents. The life cycle involves a series of molts, and the duration of development is heavily influenced by soil temperature, moisture levels, and the physiological state of the host plant.
